ABSTRACT

In Brazil, in view of the different soil and climatic conditions of the sugarcane-producing regions, a strategy of creating small breeding programs for the crop is being used. Since 2008, the sugarcane breeding program of the Federal University of Santa Maria (UFSM), has been developing new, better adapted and higher-yielding sugarcane cultivars for the climate conditions of the southern region of Brazil. Cultivar UFSM XIKA FW was developed at the Laboratories of Agroclimatology and Genetic Breeding and Plant Production of the UFSM, Campus Frederico Westphalen. It is the first mutation-induced sugarcane cultivar that was protected by the Ministry of Agriculture, Livestock and Food Supply, Brazil. The genotype used for mutation induction was cultivar IAC 87-3396, due to its good performance characteristics in the southern region. The mutation mechanism was induced in 2011, by immersing individual buds in a solution of the chemical mutagenic agent methyl methane sulfonate. The best genotypes were selected and tested in plant cane in 2013 and 2018 and in ratoon cane trials in 2014 and 2015. The new cultivar has a higher sugar production potential, and the mean yield of four years was 8.2 t ha-1 higher than that of the control, indicating a high yield potential.

ABSTRACT

Gut bacterial nitroreductases play an important role in reduction of various nitroaromatic compounds to the corresponding N-nitroso compounds, hydroxylamines or aromatic amines, most of which are carcinogenic and mutagenic agents. Inhibition of gut nitroreductases has been recognized as an attractive approach for reducing mutagen metabolites in the colon, so as to prevent colon diseases. In this study, the inhibitory effects of 55 herbal medicines against Escherichia coli(E. coli) nitroreductase (EcNfsA) were examined. Compared with other herbal extracts, Syzygium aromaticum extract showed superior inhibitory potency toward EcNfsA mediated nitrofurazone reduction. Then, the inhibitory effects of 22 major constituents in Syzygium aromaticum against EcNfsA were evaluted. Compared with other tested natural compounds, ellagic acid, corilagin, betulinic acid, oleanic acid, ursolic acid, urolithin M5 and isorhamnetin were found with strong to moderate inhibitory effect against EcNfsA, with IC50 values ranging from 0.67 to 28.98 mol·L-1. Furthermore, the inhibition kinetic analysis and docking simulation demonstrated that ellagic acid and betulinic acid potently inhibited EcNfsA (Ki < 2 μmol·L -1) in a competitively inhibitory manner, which created strong interactions with the catalytic triad of EcNfsA. In summary, our findings provide new scientific basis for explaining the anti-mutagenic activity of Syzygium aromaticum, where some newly identified EcNfsA inhibitors can be used for developing novel agents to reduce the toxicity induced by bacterial nitroreductase.

ABSTRACT

El cáncer es un problema de salud pública mundial y su incidencia sigue elevándose. El 90 a 95% de las causas se deben a factores externos, de esos factores externos, el cigarrillo representa el 20 a 25%. La Agencia Internacional para la Investigación del Cáncer (IARC) han evaluado 62 de los 5.000 compuestos químicos del tabaco o cigarrillo, siendo considerados con suficientes evidencias como carcinogénicos tanto para humanos como para animales. Entre los canceres que han sido asociados con el consumo del cigarrillo, se encuentra el Cáncer del Cuello Uterino (CaCU). El CaCu está asociado a la infección del Virus del Papiloma Humano, sin embargo, hay fuerte evidencias que el cigarrillo es un factor de riesgo independiente en la origen y evolución del CaCU.


Cancer is a worldwide public health problem and its incidence is increasing. External factors are the 90-95% of the causes, cigarette represents the 20-25%. International Agency for Cancer Research has evaluated 62 of the 5.000 cigarette or tobacco chemical compounds; they have been considered with enough evidences as carcinogenic. Among the cancers that have been associated to the cigarette or tobacco, the Cancer of the Cervix (CaC) is one of them. The CaC is associated to the infection of the Human Papillomavirus, however, there are strong evidences that support the cigarette as an independent risk factor in the origin and evolution of the CaC.

ABSTRACT

Oral cancer is the sixth most common cancer seen worldwide out of which squamous cell carcinoma accounts for 94% of oral cancer. Squamous cell carcinoma is found to be a more prevalent in south Asia because of habits like betel quid, tobacco chewing, and areca nut. Th ese along with alcohol are considered as the major risk factors. Th e mutagenic eff ects produced by these are dose and duration dependent. Some cases of squamous cell carcinoma were found in persons without habits. Early detection of oral cancer is the key factor which helps in the faster treatment and improves the survival rate of patients. A brush biopsy is the simple and inexpensive cytological technique in which an OralCDx Brush is used to collect the cells by scrubbing the mucosal site followed by spreading the sample into the glass slide which is then stained. Veloscope is a device which measures the alteration of autofl uorescence of the oral mucosa. Th e patients, in many cases, may be unaware initially as many of these lesions are asymptomatic during the initial stages and might be fi rst detected by a dentist. Th us, proper knowledge on the clinical features and eff ective use of the screening aids are essential to produce the best possible long-term outcome.

ABSTRACT

The species Citrus sinensis (L.) Osbeck (orange) is used in folk medicine as an infusion (tea) for the treatment of nerve irritation, insomnia, spasms, whooping cough, flue, and gastrointestinal problems. The aim of this study was to evaluate the antiproliferative and genotoxic potential of aqueous extracts of C. sinensis testing by Allium cepa L., as well as to determine the components in the complex mixture (tea). Leaves from two populations of this species were used in the preparation of infusions at two concentrations: 6g.L-1 and 24g.L-1, besides using distilled water as a negative control and 9.6% glyphosate as a positive control. Six groups of four bulbs of A. cepa were used, each group corresponding to one of the treatments. The bulbs were rooted in distilled water and then transferred to the respective treatments where they remained for 24 hours (the bulbs of the negative control remained in distilled water). The rootlets were collected, fixated in ethanol-acetic acid (3:1) for 24 hours and stored in 70% alcohol. 4,000 cells were analyzed per group of bulbs, and the calculated mitotic indices were submitted to statistical analysis using the c 2 test. The aqueous extracts of C. sinensis have as main components flavonoids and phenolic acids. The values of the mitotic index decreased with increasing concentration of the extracts relative to the control in water. The results indicated that the orange aqueous extracts in the studied concentrations in both populations have antiproliferative and genotoxic effects on the cell cycle of A. cepa.


A espécie Citrus sinensis (laranjeira) (L.) Osbeck é usada na medicina popular como infusão (chá) para o tratamento de irritações nervosas, insônia, espasmos, tosse convulsa, gripes e problemas gastrointestinais. O objetivo desse estudo foi avaliar o potencial antiproliferativo e genotóxico dos extratos aquosos de Citrus sinensis pelo teste de Allium cepa L., bem como determinar os componentes na mistura complexa (chá). Folhas de duas populações dessa espécie foram utilizadas no preparo das infusões em duas concentrações: 6g.L-1 e 24g.L-1, além de serem usados água destilada como controle negativo e glifosato 9,6% como controle positivo. Utilizaram-se seis grupos de quatro bulbos de A. cepa, cada grupo correspondendo a um dos tratamentos. Os bulbos foram enraizados em água destilada e então transferidos para os respectivos tratamentos onde permaneceram por 24 horas (os bulbos do controle negativo permaneceram em água destilada). As radículas foram coletadas, fixadas em etanol-ácido acético (3:1) por 24 horas e armazenadas em álcool 70%. Foram analisadas 4,000 células por grupo de bulbos, e os índices mitóticos calculados submetidos à análise estatística pelo teste c 2. Os extratos aquosos de C. sinensis têm como componentes principais flavonoides e ácidos fenólicos. Os valores dos índices mitóticos diminuíram com o aumento das concentrações dos extratos, em relação ao controle em água. Os resultados indicaram que os extratos aquosos de laranjeira nas concentrações estudadas das duas populações possuem ação antiproliferativa e genotóxica sobre o ciclo celular de A. cepa.

ABSTRACT

Degradation of chrysene, a four ringed highly carcinogenic polycyclic aromatic hydrocarbon (PAH) has been demonstrated by bacterial mixed culture Biorem-CGBD comprising Achromobacter xylosoxidans, Pseudomonas sp. and Sphingomonas sp., isolated from crude oil polluted saline sites at Bhavnagar coast, Gujarat, India. A full factorial Central Composite Design (CCD) using Response Surface Methodology (RSM) was applied to construct response surfaces, predicting 41.93% of maximum chrysene degradation with an experimental validation of 66.45% chrysene degradation on 15th day, using a combination of 0.175, 0.175 and 0.385 mL of OD600=1 inoculum of A. xylosoxidans, Pseudomonas sp. and Sphingomonas sp., respectively and a regression coefficient (R2) of 0.9485 indicating reproducibility of the experiment. It was observed that chrysene degradation can be successfully enhanced using RSM, making mixed culture Biorem-CGBD a potential bioremediation target for PAH contaminated saline sites.

ABSTRACT

At the present study, Allium cepa root tips were collected during 2,6 and 24 hrs after treated with selected concentrations (1.0,3.0,5.0 and 7.0 g/L) of Mono-Sodium Glutamate (MSG) that used as flavour enhancer in foods. MSG induced mitodepression chromosomal aberrations such as bridges, fragments, disturbance, sticky chromosomes and other morphological abnormalities like enlargement cells. In this study we amid to determine the inhibitory effect of oil extract (1.25 μl/ml) from two medical plant species Origanium majorana L. and Ruta chalepensis on this food additive. Our results demonstrated that these extracts have the potency to suppress MSG by increasing of mitotic index and reduction of the chromosomal aberration and thus could be a promising antimutagenic and antigenotoxic potential.

ABSTRACT

The compounds 6-dimethylaminopurine and cycloheximide promote the successful production of cloned mammals and have been used in the development of embryos produced by somatic cell nuclear transfer. This study investigated the effects of 6-dimethylaminopurine and cycloheximide in vitro, using the thiazolyl blue tetrazolium bromide colorimetric assay to assess cytotoxicity, the trypan blue exclusion assay to assess cell viability, the comet assay to assess genotoxicity, and the micronucleus test with cytokinesis block to test mutagenicity. In addition, the comet assay and the micronucleus test were also performed on peripheral blood cells of 54 male Swiss mice, 35 g each, to assess the effects of the compounds in vivo. The results indicated that both 6-dimethylaminopurine and cycloheximide, at the concentrations and doses tested, were cytotoxic in vitro and genotoxic and mutagenic in vitro and in vivo, altered the nuclear division index in vitro, but did not diminish cell viability in vitro. Considering that alterations in DNA play important roles in mutagenesis, carcinogenesis, and morphofunctional teratogenesis and reduce embryonic viability, this study indicated that 6-dimethylaminopurine and cycloheximide utilized in the process of mammalian cloning may be responsible for the low embryo viability commonly seen in nuclear transfer after implantation in utero.

Antecedentes: el benzo(a)pireno es un hidrocarburo aromático policíclico con efectos adversos para la salud, una de las fuentes es la ingestión de alimentos, formados durante procesamiento industrial o en el hogar. Objetivo: indagar sobre la formación de benzo(a)pireno en los alimentos, su activación biológica, relación con el cáncer, contenido en los alimentos y la normativa que regula la cantidad en alimentos para humanos. Materiales y métodos: se realizó una búsqueda bibliográfica de artículos publicados en las bases de datos nacionales e internacionales. Resultados: el benzo(a)pireno ingerido con los alimentos se absorbe por el intestino, se metaboliza predominantemente en el hígado, allí se activa y puede inducir cáncer de diversa localización, como esófago, estómago, intestino, piel, vejiga, pulmón e hígado, evidenciado en estudios experimentales en animales. El benzo(a)pireno atraviesa la placenta y es potencialmente tóxico para el feto. Las cantidades en algunos alimentos exceden las máximas permitidas por la Comisión Europea entidad que periódicamente actualiza las normas sobre el tema. En Colombia no se encontró reglamentación. Conclusión: el benzo(a)pireno procedente de alimentos genera compuestos capaces de desarrollar cáncer principalmente del tracto gastrointestinal. La Comisión Europea actualiza periódicamente la normativa que regula el contenido de benzo(a)pireno en alimentos, Colombia carece de normas sobre el tema.


Background: Benzo[a]pyrene is a polycyclic aromatic hydrocarbon which has been related with adverse health outcomes. Food is a source of benzo[a]pyrene; which is produced during industrial processing or cooking. Objective: To review information about benzo(a)pyrene formation in food, biological activation, association with cancer, food content and regulation of benzo(a)pyrene content in human food. Methods: A literature search from national and international scientific databases was developed. Results: benzo[a]pyrene ingested is absorbed by the intestine metabolized and activated, predominantly, by the liver. Animal studies have associated benzo[a]pyrene with esophagus, stomach, intestine, skin, bladder, lung, and liver cancer. Benzo[a]pyrene is potentially toxic to the fetus, due to it passes trough placenta. Benzo[a]pyrene amounts in some foods exceed the maximum level allowed by the European Commission; which periodically updates legislation on this topic. In Colombia there is not regulation about benzo[a]pyrene. Conclusion: benzo[a]pyrene in food generates compounds that may be associated with cancer, mainly gastrointestinal cancer. The European Commission regularly updates regulation about benzo[a] pyrene content in foods. Colombia does not have regulation on this topic.

The present work describes the influence of gamma irradiation (GR), ethyl methane sulphonate (EMS) and sodium azide (SA) treatment on yield and protein content of selected mutant lines of cotton. Seeds of MCU 5 and MCU 11 were exposed to gamma rays (GR), ethyl methane sulphonate (EMS) and sodium azide (SA). Lower dose of gamma irradiation (100-500 Gy), 10-50 mM EMS and SA at lower concentration effectively influences in improving the yield and protein content. Significant increase in yield (258.9 g plant-1) and protein content (18.63 mg g-1 d. wt.) as compared to parental lines was noted in M2 generations. During the subsequent field trials, number of mutant lines varied morphologically in terms of yield as well as biochemical characters such as protein. The selected mutant lines were bred true to their characters in M3 and M4 generations. The significant increase in protein content and profiles of the mutant lines with range of 10.21-18.63 mg g-1. The SDS-PAGE analysis of mutant lines revealed 9 distinct bands of different intensities with range of 26-81 kDa. The difference in intensity of bands was more (41, 50 and 58 kDa) in the mutant lines obtained from in vitro mutation than in vivo mutation. Significance of such stimulation in protein content correlated with yielding ability of the mutant lines of cotton in terms of seed weight per plant. The results confirm that in cotton it is possible to enhance the both yield and biochemical characters by in vivo and in vitro mutagenic treatments.

Abstract:The world population rising very fast and it has resulted in increasing the area of polluted soil, water and air. In order to cater to the demands of the people, the rapid expansion of industries, vehicles, etc. is necessary. But all of these developments make our world, especially urban area,heavily polluted. And many kind of contaminant substances showing bad effects to human health. Our research work aimed to evaluate Ulaanbaatar city soil contamination using sanitary microbiological and genetoxicological studies.The soils samples used in this study were collected from Ulaanbaatar city areas, from 5 points soil from the residential area of Ulaanbaatar; soil from city food market; soil from the area along the city highway; sludge of the city water treatment facilities; soil from the downtown. Genotoxicological effect of soil contaminant substances or ability to cause a mutation to living organisms was determined by Eims test and DNA damaging experiment on test strain Salmonella thyphimurium TA-100 (his G46, rfa B, bio pKm 101) and Escherichia coli (wp, uvr, pol). The result revealed that Ulaanbaatar city soil samples have inhibition effect to test strain growth rate.From our research work, we are making the following conclutions:1.Water and organic solvent solution of Ulaanbaatar soils have a high microbial growth inhibition rate. In particular, soil from the residential area has shown higher rate.2.Mutagenic activity of natural soil samples of Ulaanbaatar area was studied in bacterial test-systems. Soil samples revealed no mutagenic effect.

ABSTRACT

O extrato aquoso de erva-mate, obtido a partir de folhas secas de Ilex paraguariensis, é uma bebida amplamente consumida na América do Sul. Inicialmente, nosso objetivo foi caracterizar os compostos presentes nas amostras de erva-mate disponíveis no mercado brasileiro (CH: chimarrão; T: chá mate torrado; G: chá mate torrado, comercialmente acondicionado em garrafas ou C: em copos; TS: chá mate torrado solúvel A mutagenicidade, citotoxicidade e antimutagenicidade de todas as amostras também foram avaliadas atavés do Teste de Ames na presença e na ausência de ativação metabólica. Em seguida, analisamos a amostra TS (2,5, 5,0 e 10 mg/mL) quanto a sua atividade antioxidante e antigenotóxica. Além disso, avaliamos também os efeitos da amostra TS sobre a sinalização da leptina e da insulina no hipotálamo e o estresse oxidativo hepático de ratos adultos obesos programados pela superalimentação neonatal (S). Para induzir S, o tamanho da ninhada foi reduzido a três filhotes por lactante e as ninhadas com número padrão de filhotes (dez/lactante) foram utilizadas como controle. Aos 150 dias de vida, as proles S foram subdivididas em: TS - tratados com extrato aquoso de erva-mate (1g/kg de peso corporal/dia, por gavagem) e S - recebendo água por gavagem durante 30 dias. A prole controle (C) também recebeu água nas mesmas condições do grupo S. Em nossos resultados, verificamos a presença de ácido clorogênico, cafeína e teobromina em todas as amostras analisadas. O conteúdo de compostos fenólicos nas infusões estudadas foram CH: 5,14±0,23; T: 4,33±0,01; G: 0,93±0,25; C: 0,80±0,3 e TS: 8,35±0,5 mg/ml. Não observamos efeito mutagênico ou citotóxico nas amostras analisadas. Um efeito antimutagênico significativo foi observado para a cepa TA97 (pré-, co- e pós-tratamento), na presença de ativação metabólica, em todas as amostras testadas. A amostra TS também apresentou um efeito antimutagênico significativo para a TA102 (pré-, co-e e pós-tratamento)...


Yerba mate extract, made from dried leaves of Ilex paraguariensis, is a tea-like beverage consumed in South America. Here, we aim, firstly, to characterize the compounds present in yerba mate beverages available in the Brazilian market (chimarrao –CH; mate tea –T; mate tea commercially packed in bottles –B and cups ready-to-drink –C; and roasted yerba mate soluble tea –TS). We also evaluated its mutagenic, cytotoxic and antimutagenic properties through Ames test in the presence and absence of metabolic activation. Then, we analyzed exclusively TS sample (2.5, 5.0 and 10 mg/mL) as its antioxidant activity, on plasmid DNA cleavages and upon epithelial esophagus primary culture of Wistar rats treated with a genotoxic agent. Futhermore, we evaluated the effects of mate tea (TS) upon the hypothalamic changes of leptin and insulin signaling and hepatic oxidative stress in postnatal early overfeeding (S) rats. To induce S, litter size was reduced to three pups per dam and normal litters (ten pups/dam) were used as control. In postnatal day (PN) 150, S offspring were subdivided into: S and TS groups treated, respectively, with water or mate tea (1g/kg BW/day, by gavage) during 30 days. C offspring received water. Our results showed the presence of chlorogenic acid, caffeine and theobromine in all analyzed samples. The contents of phenolic compounds in the studied infusions were CH:5,14±0,23; T:4,33±0,01; B:0,93±0,25; C:0,80±0,3 and TS:8,35±0,5 mg/mL. No mutagenic or cytotoxic effect was observed for all analyzed samples. A significant antimutagenic effect was observed for S.typhimurium TA97 (CoT, PreT and PosT), in the presence of metabolic activation, for all tested samples. TS sample also exhibited a significant antimutagenic effect for S.typhimurium TA102 (CoT, PreT and PosT) in the presence of metabolic activation. We did not observed an antimutagenic effect using TA98, TA100 and TA102 for CH, T, C and B samples; and no effect for TA98 and TA100 for TS sample...

ABSTRACT

Foram avaliados os efeitos antiinflamatório, antibacteriano e mutagênico do extrato aquoso das sementes de Amburana cearensis. A atividade antiinflamatória foi avaliada em modelo de edema de pata induzido por carragenina, utilizando o extrato em concentrações de 10 % e 20 % nos grupos experimentais; AAS 10 mg/kg (v.o) no grupo padrão e água destilada no grupo controle. A atividade antimicrobiana foi determinada através do método de diluição em Agar, utilizando concentrações de extrato de 10 %, 7,5 %, 5 %, 2,5 % e 1 % em cepas de Sthaphylococcus aureus ATCC 27853, Escherichia coli ATCC 25922 e Pseudomonas aeruginosas ATCC 25923) e a atividade mutagênica foi determinada pelo teste de Allium cepa, utilizando extrato em concentrações de 0,02 mg/mL, 0,1 mg/mL e 0,5 mg/mL. O extrato aquoso das sementes de Amburana cearensis nas concentrações de 10 % e 20 % apresentou efeito antiedematogênico, estatisticamente significativo a partir de duas horas após administração do flogógeno, e tal efeito persistiu até 24 horas após a indução da resposta inflamatória. Quanto à atividade antibacteriana, o extrato não apresentou ação contra as cepas bacterianas de Sthaphylococcus aureus, Escherichia coli e Pseudomonas aeruginosas nas concentrações testadas. A análise dos resultados do teste de Allium cepa evidenciou ação tóxica (em concentração de 0,5 mg/mL) e mutagênica (micronúcleo 0,1 mg/mL e aberrações cromossômicas 0,1 mg/mL e 0,5 mg/mL) do extrato de Amburana cearensis em células meristemáticas de Allium cepa. Tais resultados sugerem potencial aplicação terapêutica no tratamento da inflamação. Contudo, também demonstram a necessidade de estudar para comprovar a segurança na utilização dessa espécie.


The anti-inflammatory, antibacterial and mutagenic effects of the aqueous extract of Amburana cearensis seeds were evaluated. The anti-inflammatory activity was evaluated by a paw edema model induced by carrageenan, using the extract at 10 % and 20 % concentrations in the experimental groups: AAS 10 mg/kg (orally administrated) in the standard group and distilled water in the control group. The antimicrobial activity was determined by the agar dilution method, using extract concentrations of 10 %, 7.5 %, 5 %, 2.5 % and 1% in strains of Staphylococcus aureus ATCC 27853, Escherichia coli ATCC 25922 and Pseudomonas aeruginosas ATCC 25923), and the mutagenic activity was determined by the Allium cepa test using extract concentrations of 0,02 mg/mL, 0,1 mg/mL and 0,5 mg/mL. The aqueous extract of Amburana cearensis seeds at 10 % and 20 % concentrations had an statistically significant antiedematogenic effect two hours after administering the flogogen, and this effect persisted for up to 24 hours after inducing the inflammatory response. Regarding the antibacterial activity, the extract showed no action against the bacterial strains of Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosas at the concentrations tested. The results of the Allium cepa test showed the toxic (at a concentration of 0,5 mg/mL) and mutagenicity (0.1 mg/mL micronucleus and 0.1 mg/mL and 0.5 mg/mL chromosomal aberrations) actions of the Amburana cearensis extract on meristematic cells of Allium cepa. These results suggest potential therapeutic applications to treat inflammations. However, they also show the need for further studies to demonstrate the safe use of this species.

ABSTRACT

We evaluated the safety of <i>Ashitaba</i> (<i>Angelica keiskei</i>) in bacterial reverse mutation test as well as single and 13-weeks oral toxicity tests. In the bacterial reverse mutation test, ethanol extract of <i>Ashitaba</i> had no reverse mutation inducing activity on five bacterial strains with or without S9 metabolic activation. In the single oral toxicity test, <i>Ashitaba</i> powder (3,500 mg/kg/day) showed no adverse effects in male and female SD rats. In the 13-week repeated oral toxicity test, <i>Ashitaba</i> powder (875 and 1,750 mg/kg/day) showed no adverse effects on body weight, food consumption, blood biochemistry, hematology, urinalysis, ophthalmoscopy, organ weight and histopathology in male and female SD rats. These results indicate that<i> Ashitaba</i> is very safe foodstuff under the conditions of this study.<br>

ABSTRACT

Cancer constitutes the second main mortality cause in the world, after cardiovascular diseases. In spite of the progresses in the chemotherapeutics treatments, many patients fail chemotherapy, mainly because of side effects or multi-drugs resistance, proving the need and importance of the research for new molecules with anticancer activity, more effective and with smaller adverse effects. Various compounds derived from plant secondary metabolites are commonly used in the chemotherapy against cancer and the natural products play an important role in the research for new molecules. Among several molecules of natural origin evaluated by MTT assay in murine tumor cell lines [breast (LM3) and lung (LP07)] the quinona-methide triterpenes tingenone and pristimerin showed marked cytotoxic activity presenting IC50 around 2 and 5 µM respectively. The structure-activity relationship suggests that rings A and B containing an α, ß-unsaturated carbonyl group are essential for the observed cytotoxic activity. The interaction between these positions and acetylcisteyne residues suggests a probable mechanism of action. The in vitro mutagenic activity was also evaluated by the Salmonella microsome assay (Ames test) for pristimerin and tingenone with and without metabolic activation (S9) in the strains TA98, TA97a, TA100 and TA102, none of which showed mutagenic potential in any strains. Estrogenic and anti-estrogenic activities were also studied by the e-screen assay in MCF-7 cells with negative results. The present data point to the importance of pristimerin and tingenone as representative of an emerging class of potential anticancer chemicals.

ABSTRACT

The purpose of the present investigations was to improve the yield of alkaline protease for leather dehairing by subjecting the indigenous proteolytic strain Bacillus licheniformis N-2 to various mutagenic treatments viz. UV irradiations, NTG (N-methyl-N-nitro-N-nitrosoguinidine) and MMS (methyl methane sulfonate). After screening on skim milk agar plates, a total of nine positive mutants were selected for shake flask experiments. Among these, the best proteolytic mutant designated as UV-9 showed 1.4 fold higher alkaline protease activity in preoptimized growth medium than the parent strain. The fermentation profile and kinetic parameters such u(h-1), Yp/s, Yp/x, Yx/s, q s, Qs, q p and Qp also indicated the superiority of the selected mutant UV-9 for alkaline protease production over the parent strain and rest of the mutants. The dehairing capability of mutant UV-9 alkaline protease was analyzed by soaking goat skin pieces for different time intervals (3-15 h) at 40 º C. A complete dehairing without degradation of collagen was achieved after 12 h, indicating its commercial exploitation in leather industry.

ABSTRACT

Pueraria mirifica is a Thai phytoestrogen-rich herb traditionally used for the treatment of menopausal symptoms. Pueraria lobata is also a phytoestrogen-rich herb traditionally used in Japan, Korea and China for the treatment of hypertension and alcoholism. We evaluated the mutagenic and antimutagenic activity of the two plant extracts using the Ames test preincubation method plus or minus the rat liver mixture S9 for metabolic activation using Salmonella typhimurium strains TA98 and TA100 as indicator strains. The cytotoxicity of the two extracts to the two S. typhimurium indicators was evaluated before the mutagenic and antimutagenic tests. Both extracts at a final concentration of 2.5, 5, 10, or 20 mg/plate exhibited only mild cytotoxic effects. The plant extracts at the concentrations of 2.5, 5 and 10 mg/plate in the presence and absence of the S9 mixture were negative in the mutagenic Ames test. In contrast, both extracts were positive in the antimutagenic Ames test towards either one or both of the tested mutagens 2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide and benzo(a)pyrene. The absence of mutagenic and the presence of anti-mutagenic activities of the two plant extracts were confirmed in rec-assays and further supported by a micronucleus test where both plant extracts at doses up to 300 mg/kg body weight (equivalent to 16 g/kg body weight plant tuberous powder) failed to exhibit significant micronucleus formation in rats. The tests confirmed the non-mutagenic but reasonably antimutagenic activities of the two plant extracts, supporting their current use as safe dietary supplements and cosmetics.

ABSTRACT

Do material recolhido por aspiração no interior de uma residência no Rio de Janeiro, com uso de aspirador de pó, por 4 horas contínuas de captura, em todos os cômodos da casa, foram encontrados 1.284 Rhipicephalus sanguineus, dos quais 289 eram fêmeas e, destas, uma tinha malformação no opistossoma que estava assimétrico, e monstruosidade pela presença de dois ânus. Esse é o primeiro registro desse fenômeno em R. sanguineus.


From material collected by into a residence house at Rio de Janeiro city, using an aspirator machine during four continues hours in all house, it was captured 1,284 Rhipicephalus sanguineus including 289 females; one of them showed a mutagenic alteration like an asymmetry of opistosome, and monstrosity like a two anus. The first register of this phenomenon in R. sanguineus is reported.

ABSTRACT

We report a case of AML-M1 with 5q aberration at diagnosis. The patient was treated with high-dose chemotherapy (HDCT). After remission induction, he received allogenic peripheral blood stem cell transplantation (PBSCT) from an HLA-match donor brother. The successive follow-up conventional cytogenetics investigations in remission after HDCT and PBSCT revealed cytogenetic remission. The most interesting observation in this case is that relapsed marrow revealed the emergence of an entirely new, highly aberrant, unrelated clone with unusual translocations t(6;17)(p23;p11.2),+8,der(8)dup inv(8)(q23qter), t(10;19)(q26;q13.3) 4½ months after PBSCT. Our findings suggest the possibility of a mutagenic effect of HDCT and myeloablative intense chemotherapy before PBSCT that could have induced a genetic lesion in the recipient's genetically unstable stem cells in an environment of immunosuppression. The highly complex nature of the clone and the rapid clonal evolution indicates the possibility of selective pressure with proliferative advantage.

ABSTRACT

<p><b>OBJECTIVES</b>It is important to assess the risk of static magnetic fields (SMFs) on human health, because epidemiological studies have indicated that SMFs play a role in the development of diseases such as leukemia and brain tumor. In our environment, we have numerous chances to be exposed to not only SMFs but also many chemicals containing mutagens. The aim of this study is to investigate the effect of SMFs on the induction of micronuclei induced by some mutagens.</p><p><b>METHODS</b>BALB/c mice were exposed to 4.7 tesla (T) SMF for 24 hr immediately after the injection of carboquone (alkylating agent), colcemid (spindle poison), mitomycin C (cross-linking agent), vincristine (spindle poison), sodium fluoride (a byproduct of aluminum plants under strong SMF) or 1-ethyl-1-nitrosourea (brain tumor-, gliomas- and thymic lymphoma-inducing chemical).</p><p><b>RESULTS</b>The frequency of micronuclei induced by six mutagens increased after co-exposure to SMF.</p><p><b>CONCLUSIONS</b>An additive/synergistic effect of SMF and chemicals was observed from the results of increased frequency of micronuclei induced by mutagens in mouse bone marrow erythrocytes.</p>
